- 0:06Welcome back to another episode of the Biohop podcast.
- 0:09Today I'm thrilled to be joined by Lee Fang, who is the Co
- 0:12founder and CEO of Excalapoint. In today's episode, we're going
- 0:17to talk a little about Lee's career and the work that
- 0:19Excalapoint are doing in the T cell engager space.
- 0:22Lee, thank you so much for coming on and joining us.
- 0:26Hi, George. It's very appreciate to give us
- 0:29the opportunity to talk about our story and our science and
- 0:33what do we do. So I'm Leifang and you can call
- 0:38me late. I was trained in Chinese kind of
- 0:42a science as AT cell immunologist and which is a
- 0:47little bit different from my classmate.
- 0:51I think my career which is just the start, finished my PhD
- 0:55training and direct go to the industrial.
- 0:58I first, my first job is to join the GSK and the China and
- 1:03further pursue my research on T cell differentiations in
- 1:08autoimmune conditions. But it's from the different
- 1:10angles. It's for the drug development
- 1:12with screening small molecules which could be suppress the T
- 1:17cell activities and we will promote the T activities.
- 1:21We want to control, control the autoimmune progressions in
- 1:27multiple sclerosis, which is neuroinflammation disease and
- 1:31also rheumatoid psoriatics with, you know, with the outing
- 1:36inflammations in the in the joint and after in GSK for
- 1:41almost 7 years, which I really learned how to develop an
- 1:46innovative drugs in the in the outing settings.
- 1:49So back to the 2015, I got an opportunity to as a entrepreneur
- 1:56to sell biotech in China and the starting from the the companies
- 2:01called IMAP and that's really fantastic opportunities because
- 2:072015 is the starting of you know the immunotherapies year because
- 2:14the approval for PD1 antibodies such as Keytruda and Optivo.
- 2:19So we worked as a map at that time.
- 2:21We're focusing on the new generation e-mail checkpoint
- 2:26development for example, like in CD37, like in CD 73, we are
- 2:31seeking for another PD one on the road for the development.
- 2:35Beside of that I think it is more related to today's we come
- 2:40back to the 2015 we're starting for the bispecific development
- 2:45and here our initial rationale is that there were no bispecific
- 2:49e-mail checkpoint combination will have to resolve the re
- 2:55refractory or UN responder for PD1 immune therapies.
- 3:00There are developed the first time I I joined the Tsung
- 3:06engaged development based on the full one BB based bispecific.
- 3:11Yeah, IMF there at MMAP and ABL bio now it's a a Co stock list.
- 3:18The company together we developed the four one BB based
- 3:22platforms. Another I think it is more
- 3:25related today is calling based 41 BB.
- 3:28At that time, we find the four one BB based bispecific can
- 3:32solve problems of four one BB, which in the past the four one
- 3:37BB has shown the very severe liver tacticities and to which
- 3:43can block its further explorations on ultimate
- 3:47condition in in the liver with the severe liver tox and.
- 3:53However, by using the bispecific and we can precisely control the
- 3:59activation of four MBB which only happens on the in the tumor
- 4:05because one only binds one arm bind to the tumor the other by
- 4:11casting and they then for one they become deactivated.
- 4:15So we can precisely control the the tissue activation which is
- 4:19far from the liver. So in that the case, you can
- 4:22make the in the tumor the four one BB activated and in the
- 4:28without liver toxicity. So that is very smart designs
- 4:32and recently these molecules we call them Chiva show very good
- 4:39advocacies in the in the gastric cancers in the first one and
- 4:44which come back to the 2020 we where M AB was listed on the
- 4:50NASDAQ. I just wanted to pursue a new
- 4:53career at that time the chairman of Lupu Bio Pharma, which is
- 4:58listed on the Hong Kong exchange invite me to join the Lupu as a
- 5:04head of research. So that's easier.
- 5:09I joined Liverpool as a as a private space and as a senior
- 5:14leaders, we lead the Liverpool biopharma list in Hong Kong.
- 5:18And before, before I leave Liverpool 2020, before I leave
- 5:26Liverpool 2025, they become a really biopharma company in
- 5:31China. So I look at the local
- 5:35biopharma. I also participate in the lead a
- 5:38lot of ADC development. And we I also drive the two
- 5:47person class ADC, one is TPC, three ADC and another is the Cdr
- 5:5317. Basically ADC move, bring them
- 5:57to the to the clinic and then in parallel for to further pursue
- 6:03my interest in the career in the T cell engagers.
- 6:07I find that in the four one BB, there's some limitations when I
- 6:10go for the more with cosmic cold tumor, that means there's almost
- 6:16no T cell infiltration situations.
- 6:18The four one BB doesn't work well.
- 6:20So and the further explorations we find when you bring the CD 3
- 6:25and the four one BB together, we can do it.
- 6:28So I incubate that platforms in the Liverpool biopharma and
- 6:33which is likely which come back to the end of 2024.
- 6:39We have opportunities because the the local biopharma wanted
- 6:44more focus on their development of ADC and because there is one
- 6:49PD one approving China, the second is a eufr based ADC
- 6:54already approved in China, one or two focusing ADC and and the
- 6:59PD 1. So I have an opportunity to
- 7:02taking out the TCe and related the TCe platform out.
- 7:10So at that time I call for my partner Jalen.
- 7:13I just a call, I know Jalen, we know, I know Jalen is when IMAP,
- 7:19he was the first CFO of IMAP and bring the I'm up to the next to
- 7:26the NASDAQ. So I give him a call.
- 7:29We have a hot pot dinner. So and then we decided to do it
- 7:32together. So we are lucky ones when they
- 7:37get to the opportunity to to to get a financing and also to
- 7:42bring the leading asset out and also related to CD3's platforms
- 7:49out. And there is where started from
- 7:52Excel point. We finish that deals about $41
- 7:57million financing at the August of 2025 S after very quick you
- 8:07know, you know, development of our progress of our of our
- 8:13programs and also our our platforms.
- 8:16So we get a very quick follow on after six months later, so
- 8:20almost at the end of 2024 earlier 2026 because pass around
- 8:28it's about $27.7 million. So it's in total it's almost
- 8:37reaching the six $8 million. So we have, you know, very good
- 8:43positions to further develop the T cell engages in solid tumors
- 8:49and in more specific autoimmune conditions.
- 8:53So that's where the startings of Excel point.
- 8:57Fantastic. Thank you.
- 8:58Thank you for that overview. And yeah, like you said, you,
- 9:01you launched with, you know, one of the largest seed financing
- 9:04for for Chinese biotech for people that are less familiar
- 9:08with with the company. Talk us through Excalar Point
- 9:11Science in a little bit more detail.
- 9:13When what gap did you believe existed in the T T cell engager
- 9:17and immunology, you know landscape that you know made you
- 9:20really start Excalar point? So that is for the T cell
- 9:24engagers is a long journey. It's not just what something
- 9:28started the 2010 file which come back to the 2014 I mean almost
- 9:34more than 10 years ago. It's the first T cell engagers
- 9:37Blencia only the CD 19 CDC have been approved, but it is going a
- 9:42long way until the 20 222027 a first wave of T sign engagers
- 9:51have been approved, which is almost on the hemidopole cancer
- 9:54like in CD20 PCMA related T sign gauge approval in in DLPCR and
- 10:02etcetera. However and the I think the
- 10:07milestone event was the the first half of 2024 that the
- 10:12first DL3 CD three that is the first T cell engages have been
- 10:19approved in solid tumors. I think for the T cell
- 10:23immunotherapy that's a real milestones for several reasons.
- 10:26For the first one that is the first immune therapies have been
- 10:30approved. The efficacious that in the past
- 10:35was PD 1, you know, not less effective tumor site and the
- 10:42first immunotherapy approved under PD1 relapse conditions
- 10:47because you know there's no e-mail service have been
- 10:50demonstrated after PB 1. And I think that the second
- 10:55thing is that this trial is this is a benchmark.
- 11:00Tell us, you know, what's the you know, the features of
- 11:05functional design gaugers that is, you know, the new need of
- 11:11first clean tumor social antigens and there is there is,
- 11:16you know, this applications profile is like in the response
- 11:20rate is now sold as high as at ADC.
- 11:24You know, 50 or 7060% of the response rate is only 40% of
- 11:30response rate. However, the peer the OS benefit
- 11:33is very, very significant almost reach 14 months.
- 11:38So moderate response rate and KB can be applications after PD1
- 11:45and you know, very long OS. So this is set up a very good
- 11:49standards and a good, you know, role models how he saw engaging
- 11:54solid. However, currently you can see
- 11:56the car teeth not going so well, but the T cell engagers coming
- 12:01coming out. So that's I think the the that
- 12:05really draw a lot of attentions by the investors by whole field
- 12:10to know it's time for solid tuners.
- 12:12I think for us is, is that we wanted to, you know, to to we
- 12:18can do better. What's the next making the
- 12:22better T sound engagers when coming to the CD 3 alone.
- 12:26And I think another milestone that is because recently for the
- 12:32T sound engagers field move into immune reset settings by using T
- 12:38cell engages in autoimmune conditions.
- 12:40We see a lot of you know, car tease.
- 12:43We this will also happen on the 2024.
- 12:45So peace allow this. That's trials that's cases.
- 12:50We will let us know the T cell engages can be, you know,
- 12:53working on autoimmune conditions, making a whole
- 12:56immune reset. That means the research of holy
- 13:01B cell immune reptile by making self reactive base off into
- 13:08truly naive and and for second of cure, you know for cure for
- 13:14six months or even longer. So that is also tremendous.
- 13:18I think that's why T sign engagers do try draw a lot of
- 13:23attentions on that for external point for the solid tumors.
- 13:27We wanted to expand the we have pipelines to expand T sign
- 13:32engagers not only for small salon, we want to we have
- 13:36expansion to the Pine GI cancers is our second pipelines.
- 13:41We want to make him into the clinic at the end of this year.
- 13:47And then we also have a prostate cancers.
- 13:49We also have you know, even though non small cell lung
- 13:52cancers in the past, we're making T sign get can benefit
- 13:56more. But the different the things are
- 13:59different, it's more difficult that is, you know, the different
- 14:02indications the you'll need a different kind of T cell
- 14:05engagers. We wanted we wanted to not only
- 14:09CD3 we under the CD3 plus. So we added the Co Stimu signals
- 14:14as a as a full one BB to which could benefit the with the very
- 14:20few that T cell efficiency in the cultural situations like in
- 14:24CRC like in like even cold population in the small salon
- 14:30and also in the past the steep, you know, prostate cancers is
- 14:36supposed to be very cold tumor type.
- 14:38The PD one even not proving these indications.
- 14:42And that's our visions for for for the tissue and getting solid
- 14:50tumors and how another thing is for the autoimmune conditions.
- 14:55Our rationale is a little bit in the past they, they do the whole
- 15:02piece of depletion and even reset for the new one.
- 15:06And we wanted to be a more precisely to depletion only
- 15:11pathogenic results. We wanted to precisely control
- 15:16yet in that in that situations we can move the piece of
- 15:21depletion strategy to other less severe autoimmune conditions for
- 15:27you like maybe Sjogren's syndrome, maybe other you know,
- 15:31B cell related autoimmune diseases not use lisal or lab
- 15:38threaten like in SLE, like in terminal s s system siculosis.
- 15:44This kind of indications we want to just move to using only
- 15:48depletion for pathogenic B cells.
- 15:51So that's this is making Excel point could, could, could be
- 15:58differentiate. We wanted to to be
- 16:00differentiated in the ultimate and and and solid humours.
- 16:08No, absolutely. And since you've launched as
- 16:11well you've you know, rapidly built these platforms,
- 16:15progressed the six programs in a, you know, very short amount
- 16:18of time. What's being key for you to
- 16:20executing, you know at that speed and you know, while making
- 16:24sure the quality and decision making has been so effective?
- 16:28I think the based on three aspects.
- 16:32The first aspect I think for the I just I just mentioned that the
- 16:37T cell immune therapy as ATCT cell engager as a so T cell
- 16:43immune therapy in the in autoimmune for the science part
- 16:46is having demonstrate we don't worry about science.
- 16:49I just we just need to better precisely control how you
- 16:53control the T cell and to kill a target and make it more clean.
- 16:58So what we need is for engineering, good engineering,
- 17:04it's not easy. So the first you have the
- 17:09certain the road is clear, you just need to the T cells.
- 17:13I think the second is we have very good basis because we have
- 17:20you know a lot of molecular tools like in we have ACD 3
- 17:24sequencing ready, we have US coasting molecules or they're
- 17:28already ready like in for one BB even though we have pursue other
- 17:33costing like in CD 2. So in that case, because the Co
- 17:38rationale that I called my first principle, first principle of T
- 17:42sign engager field is how to using multi specific antibodies
- 17:48to reconstitute immunologic synapse as natural as we could.
- 17:54So you can you can you go when you zoom in the key molecules in
- 18:00the immunologic synapse, there's a CD3, there is a cosine for one
- 18:06BB we have there's just CD2 also we have this is it.
- 18:11That means the answer is also you need you how to dissolve
- 18:16resolve the problems in by protein engineer.
- 18:21So that's only engineering things.
- 18:23I think for the shoulder layer, you should worry understanding
- 18:30the biology of the you know, the indications.
- 18:35We we explore the different indications based on the
- 18:38different indications on the on the T cells.
- 18:41We apply the different you know, sometimes we're using for one
- 18:45baby, sometimes we're using CD2 is based on dependent tumor T
- 18:50cell in condition for using that.
- 18:53And for the even though I think for the for the for the B cell
- 18:57depletions, we only apply to CD3.
- 19:02We didn't, we didn't using cocin because we just wanted to T cell
- 19:06kill the B cell and we don't want to persist the T cell
- 19:12activated because you know, ultimate conditions you you kill
- 19:15the B cell, but you don't want to promote the T cell to other
- 19:18ultimate conditions. That's that's why we quit
- 19:22because we all the molecule to molecular tubes, we are already
- 19:25enhanced and we based on the different indications we tailor
- 19:30made the different, you know, molecules on that.
- 19:34I think for the third all because all these things in the
- 19:39clinic, we don't know we, but we have the the luxury because in
- 19:46in we can apply both the in China and the US clinic
- 19:50resources. It's possibly in the China there
- 19:53are huge a mathematically large patient population needed to be
- 19:58treated. So in that case, we can do quick
- 20:02clinical to see we can come for 10 people or tons of people.
- 20:08Patient data to tell us whether is this design is right or not
- 20:14because in the past we already developed a very deep know how
- 20:19to molecule design and how this performance in the Sino.
- 20:23Currently, we also have initially clinical radar.
- 20:25We we build up, you know, this kind of algorithm, but this is
- 20:30in mind human algorithm to to build to quick build up, you
- 20:34know, T sign engaged in the clinic.
- 20:36So for the second program server program, we are more quick
- 20:40because we have that. And I think for the last
- 20:43currently we are also working with AI with agents.
- 20:49We wanted to translate our knowledge of the TESAR engager
- 20:54to to to to AI and then AI back to help us to more quick to
- 21:02design the next generation T sound engagers.
- 21:05Before example, I will give you example our first T sound
- 21:08engagers. We almost spend three years to
- 21:13make the molecules in the clinic based on our definitely know how
- 21:19can we only use 11 months to make our prostate cancer
- 21:29programs into the clinic. We shorten because we have two
- 21:33ready and we have you know how to design and we get the quick
- 21:38answers from the Sino data and isn't got the quick design.
- 21:43We know which range that could be successful in the human.
- 21:47I think maybe in the future by using AI, maybe we can get more
- 21:52quick design on the T sign engagers based on that one.
- 21:55That's why I think I have a confidence excellent point.
- 21:59Make a better drive in the in the clinic, yeah.
- 22:02Now that makes sense and leave built the company up, you know,
- 22:05very quickly. You've got funding behind you
- 22:07just quickly. Before we finish, what's the
- 22:09plan now for you and Excalar Point over the next 12 months?
- 22:14That is because of the very well we're very looking forward we
- 22:20would we have we could achieve the first.
- 22:23We just wanted to further just explore what our initial TSR
- 22:30engaged CD3 Co team design, what's the safety and what's the
- 22:35potential efficacious and what's the dose in the human as the
- 22:39first things. And from that clinical data, we
- 22:42can really learn a lot is we, because we are the first one to
- 22:48design the CD 3 and the costing for one BB in C-section.
- 22:53That means in the working on simultaneous one because in the
- 22:58past, a lot of people working on in the combo because two
- 23:01bispecific signals in the combo situations you sometimes we they
- 23:06don't have other smart designs. They, they, they bind to CV3,
- 23:10but not for one baby. The bind for one be done not CV
- 23:13three. You are you are regional combo,
- 23:16but we are the first one to design they're working together,
- 23:19which more mimic the natural pizza activations.
- 23:23And I think this is the first one to to try them in human and
- 23:28we would we can learn that and we will accelerate our next, you
- 23:34know, new acting CD to new molecule design because we we
- 23:38learn we could achieve this. This kind of design can be
- 23:42potential safety may even though that that how is performance in
- 23:48human. It's what's what I'm really
- 23:49looking for what happened on the next we are also moving forward
- 23:57you know to push our prostate cancer and also our precisely be
- 24:03a pathogenic piece of depletion approach.
- 24:06I think maybe we can expect to these molecules into clinic
- 24:12maybe at the second-half of 2027 in that we will have more
- 24:18answers on our you know waiting molecule design and you know
- 24:22which can help on the patient worldwidely.
- 24:27Fantastic. Look, it's been a great story.
- 24:31Thank you so much for sharing a little bit about your career and
- 24:34everything around Escala Point. Everyone listening is going to
- 24:37be a really excited watching you guys over the next 12 months.
- 24:40So, Lee, thank you so much for coming on the podcast.
- 24:44And I look forward to, you know, seeing how everything goes over
- 24:47the next few few months and yeah, later on in the year.
- 24:51Yeah, thanks. It's a great talk.
- 24:53And then we wanted to share more stories in the future.
- 24:56Yeah, absolutely. Thank you, Lee.
