- 0:30with Ben Lamb. Ben Lamb the CEO and Co founder
- 0:33of Colossal. That was JRE episode hashtag 23

- 0:56extinction? It seems like quite a.
- 0:58Well, it really boiled down to his curiosity and then this key

- 1:03George Church from Harvard, the father of synthetic biology.
- 1:07That's the one apparently quite tall, and Lamb mentioned he has
- 1:10narcolepsy too. Just an interesting detail about
- 1:12this brilliant guy. So Lam's background wasn't in
- 1:16biology, was it? It was more software building
- 1:18teams. Exactly.
- 1:20Software assembling, groups of really smart people.
- 1:23He was initially interested in synthetic biology more broadly,
- 1:26like for crops, animals, that sort of thing.
- 1:29Then he gets on this call with George Church.
- 1:31Lam's firing off questions about synthetic biology.
- 1:34And church is keeping up, presumably.
- 1:36Oh yeah, Then apparently Church just sort of casually drops in
- 1:40that he's working on mammoths. On that bringing back woolly
- 1:43mammoths just like that. Pretty much.
- 1:45And Lamb's reaction, understandably, was disbelief.
- 1:48Like, wait, what? Even coming from church.
- 1:50I can imagine. So what did he do?
- 1:52What any curious person would I guess he dove into research
- 1:56overnight, looked into Church's work, and found this consistent
- 1:59mammoth through line, as he called it.
- 2:02So it wasn't just a random comment search.
- 2:04Have been working on this for a while.
- 2:06For years, and get this within seven days of that phone call,
- 2:12Lamb was in church's lab. A week from wait Mammoths to
- 2:17Let's Start a company. Yep, chose Lamb's drive right
- 2:20And how compelling Church's vision must have been.
- 2:23That was basically the start of Colossal.
- 2:25Wow. OK, so the woolly mammoth was
- 2:27the first big target. Makes sense if Church already
- 2:30had years invested. Eight years of work.
- 2:32Yeah. Plus there is this strong
- 2:34ecological argument. For bringing them back.
- 2:36Ecological modelling suggested to that reintroducing mammoths,
- 2:39these large herbivores, could actually benefit Arctic
- 2:42ecosystems, help restore the mammoth step, maybe even slow
- 2:46permafrost thaw. Interesting.
- 2:47So it wasn't just about the spectacle.
- 2:49No, not at all. And crucially, the technologies
- 2:52they needed to develop Advanced gene editing, ancient DNA
- 2:55recovery, maybe artificial wombs down the line.
- 2:58These have huge potential applications elsewhere.
- 3:00Helping existing elephant populations.
- 3:02Exactly, elephant conservation is a big one and even
- 3:05potentially spinning off tech for human healthcare.
- 3:08Right. And then the company went
- 3:09public, got a lot of attention. It went viral, basically, and
- 3:13that led to other researchers approaching them, people working
- 3:16on things like the thylacine, the dodo.
- 3:18So the scope expanded pretty quickly.
- 3:20It really did. OK, let's dig into the actual
- 3:23process. How do you even start trying to
- 3:26bring back a mammoth? The DNA must be the first
- 3:29hurdle. Oh, absolutely.
- 3:31Ancient DNA is well, Lamb called it a shitty jigsaw puzzle.
- 3:36I like that because it's so degraded.
- 3:39Totally. It's fragmented.
- 3:40It's contaminated with bacteria, fungus, whatever is in the soil.
- 3:44It breaks down really fast. So putting it back together.
- 3:47It's primarily an AI and compute problem, he emphasized.
- 3:51You need sophisticated algorithms to sort through all
- 3:54that mess, identify the real mammoth bits and figure out how
- 3:57they fit. OK, so you've got these
- 3:59fragments, then what? You compare it to a living
- 4:02relative, right? Genotyping is key, and this is
- 4:04where it gets really interesting.
- 4:06The closest living relative to the woolly mammoth isn't the
- 4:09African elephant as you might assume.
- 4:10No, it's the Asian elephant. They share something like 99.6%
- 4:15of their DNA. Wow. 99.6% of their DNA.
- 4:18Wow, 99.6% closer than Asian elephants are to African
- 4:22elephant. Exactly.
- 4:24Which is a pretty surprising fact, and it suggests you know
- 4:27that the basic cellular machinery is probably
- 4:30compatible. But that .4% difference, yeah,
- 4:33it's still a lot of genetic information, right?
- 4:35That's what makes a mammoth a mammoth.
- 4:36It is. It contains the genes for cold
- 4:38adaptation, the shaggy coat, the small ears, the fat layers, all
- 4:42that stuff. So editing needs to be precise.
- 4:45And when did they actually go extinct?
- 4:47It feels like ancient history, but.
- 4:48Well, the mainland populations died out maybe 10,000 years ago,
- 4:52but there was this isolated group on Wrangel Island off
- 4:55Siberia. And they survive longer.
- 4:58Yeah, until about 4000 years. AGO 4000.
- 5:00Wait, that means they were still around when the Egyptians were
- 5:04building the pyramids? Mind blowing isn't it?
- 5:06Mammoths and pyramids overlapping in time.
- 5:09That really changes the picture. So what happened on Rangel
- 5:11Island? The theory is basically a
- 5:13genetic bottleneck. Small population inbreeding,
- 5:17loss of genetic diversity. Eventually they just couldn't
- 5:20cope. A sad end.
- 5:22A cautionary tale, and Lam mentioned there isn't some big
- 5:25global backup of DNA for all species, right?
- 5:28No, apparently not. No comprehensive Noah's Ark bio
- 5:31vault as he put it. Which is why Colossal is having
- 5:34to build these reference genomes themselves for the species they
- 5:38work on. It's a huge undertaking.
- 5:40OK, so you have the fragmented ancient DNA, you have the Asian
- 5:43elephant genome as a reference. How do you bridge that .4% gap?
- 5:49That's the synthetic biology part.
- 5:50Exactly. This is where tools like CRISPR
- 5:53come in the. Gene editing technology.
- 5:55Right. Think of it like precise
- 5:56molecular scissors. You use the ancient mammoth DNA
- 6:00as a guide to make specific edits in the Asian elephant
- 6:03genome. You're adding back those key
- 6:05mammoth genes. So you're not aiming for a 100%
- 6:08perfect genetic match to the ancient mammoth?
- 6:10No, not necessarily. Lamb talked about functional de
- 6:13extinction. The goal is to create an animal
- 6:15that looks like a mammoth, acts like a mammoth, and most
- 6:18importantly, can perform the ecological role of a mammoth.
- 6:21Gotcha. OK, so before they jump straight
- 6:23to making mammoths, they needed a proof of concept, the woolly
- 6:26mouse. Yeah, the Woolly Mouse Project.
- 6:28It sounds a bit funny, but it was scientifically important.
- 6:30Why mice? They're pretty far from
- 6:32elephants, evolutionarily. Exactly about 200 million years
- 6:36of divergent. So just sticking mammoth genes
- 6:40directly into a mouse embryo probably wouldn't work well and
- 6:42raises ethical questions about potential harm, right?
- 6:45So instead they identified the mouse equivalent genes related
- 6:49to hair growth and phenotype the physical characteristics of the
- 6:53hair. And edited those to make them
- 6:55more. Woolly precisely trying to mimic
- 6:58the effect of the mammoth genes, but using the mouse's own
- 7:01genetic architecture. It was a safer, more targeted
- 7:04way to test the editing technology.
- 7:06And how did that go? Surprisingly, viral people love
- 7:09the idea of woolly mice. There were T-shirts, apparently
- 7:12even a meme coin. Seriously.
- 7:14Yeah, but scientifically, it was a big success.
- 7:17They created 36 healthy woolly mice. 36 and healthy.
- 7:22And crucially, it demonstrated 100% efficiency in Multiplex
- 7:26gene editing. Multiplex, meaning editing
- 7:28multiple genes at once. Exactly, editing several genes
- 7:31simultaneously and importantly with no detectable off target
- 7:34effects, no unintended changes elsewhere in the genome.
- 7:38That's a huge deal for the safety and precision of this
- 7:40tech. Wow, OK, that's a significant
- 7:43step. Did the mice give any clues
- 7:46about what mammoths actually looked like?
- 7:48Well, the woolly mice ended up having the sort of golden brown
- 7:51fur. Golden brown.
- 7:52Yeah, which apparently aligns pretty well with some of the
- 7:54well preserved mammoth carcasses found in permafrost, like Liuba,
- 7:58the baby mammoth. So it offered a visual
- 8:01confirmation too. OK, so Colossal isn't just about
- 8:04bringing back extinct giants. They're also involved in
- 8:06conservation Now, working with various groups.
- 8:09Very much so, Lamb emphasized. Working with private land
- 8:13owners, governments, indigenous people.
- 8:16It's not just happening in a lab vacuum.
- 8:18Right, he mentioned a bison project.
- 8:19Yeah, they created a population genomics map for bison,
- 8:24understanding the genetic diversity in history within the
- 8:27bison population. And that led to conversations
- 8:29with the MHA Nation, Mandan, Hidatsa and Arakara Nation.
- 8:33Exactly. And the feedback from the MHA
- 8:35Nation wasn't just about bison. They highlighted the importance
- 8:39of wolf conservation. Eagles fish a whole ecosystem
- 8:43approach. And they mentioned a specific
- 8:45wolf. The great white wolf, which
- 8:47holds cultural significance for them and their ancestral
- 8:50knowledge, even hinted at a possible connection to the
- 8:53direwolf. The direwolf like from Game of
- 8:55Thrones. The very same, though the real
- 8:57ones were distinct from Gray wolves.
- 8:59But Lamb saw this as a fascinating link and also a way
- 9:02to engage a whole different audience.
- 9:04Yeah. Sci-fi and fantasy fans, kids.
- 9:06Yeah, it definitely has broader appeal.
- 9:09But finding direwolf DNA was tough, wasn't it?
- 9:11Most fossils are from the La Brea Tar Pits in LA.
- 9:14Right. And tar isn't great for
- 9:16preserving DNA. Too hot, Too chemically harsh.
- 9:19But they did find some elsewhere.
- 9:21They managed to sequence DNA from a 13,000 year old tooth
- 9:24found in a cave. OK, how good was the sequence?
- 9:27About .15 X coverage, so not great enough to learn some
- 9:31things, but not really enough to attempt de extinction.
- 9:34That means they only got fragments covering about 15% of
- 9:36the genome. Not enough pieces of the puzzle.
- 9:38Exactly. But then they got lucky.
- 9:41How so? A much older skull, 72,000 or
- 9:4473,000 years old, was found in a riverbed somewhere.
- 9:48Older but better preserved. Yes, because it contained the
- 9:51Petrus bone. The inner ear bone I've heard
- 9:53that's incredibly dense. Super dense, it protects DNA
- 9:57exceptionally well over long periods.
- 9:59And from that bone they got 13 to 14 X coverage. 13 times
- 10:04coverage. That's way better A.
- 10:05Game changer, and enough overlapping fragments to piece
- 10:09together a much, much more complete picture of the dire
- 10:12wolf genome. Enough potentially to actually
- 10:15bring it back. Wow, OK, that's huge.
- 10:17Now what about the ethics? The whole playing God argument
- 10:21comes up a lot. It does.
- 10:22Lamb addressed it directly. His perspective is basically
- 10:25that humans are already profoundly impacting the planet.
- 10:28Over fishing, over hunting, deforestation, climate change.
- 10:32We've driven species extinct. So using technology to try and
- 10:35undo some of that damage is. Arguably a more responsible path
- 10:39than just letting ecosystems continue to degrade.
- 10:41He pointed to the successful wolf reintroduction in
- 10:43Yellowstone. Right.
- 10:45That had massive positive ripple effects through the whole
- 10:47ecosystem. Exactly, bringing back a
- 10:49keystone species can and revitalize a landscape.
- 10:51There's also this philosophical debate, isn't there?
- 10:54What even is a species? Is it about breeding, looks?
- 10:57Genetics. It's complex.
- 10:59The definition varies. Can they interbreed and produce
- 11:02fertile offspring? Do they look similar?
- 11:05Morphology. Are they genetically distinct?
- 11:07Are they geographically isolated?
- 11:09Scientists don't always agree. So even if Colossal creates
- 11:13something that looks like a mammoth and has mammoth genes,
- 11:16is it really a mammoth? Some paleontologists and
- 11:20scientists might debate that. Yeah, especially if the
- 11:23morphology, say, the tooth structure isn't perfectly
- 11:26identical even if the genes are there.
- 11:28It gets back to that Jurassic Park idea.
- 11:30Are they creating genetically modified elephants or truly
- 11:34bringing back mammoths? Right Lamb leans towards
- 11:37functional de extinction if it serves the ecological purpose.
- 11:39If it fills that niche, maybe the precise label matters less
- 11:43than the positive impact. Makes sense.
- 11:45OK, shifting gears slightly, Colossal isn't just focused on
- 11:48extinct animals, right? They're actively involved with
- 11:51endangered species now. Yes, and that's a really
- 11:53important part of their mission. They paired the extinction
- 11:56projects with species preservation, and they plan to
- 11:58open source their conservation technologies like.
- 12:00With the red wolf. Exactly.
- 12:03The red wolf situation is dire. Critically endangered?
- 12:06Maybe only 15 left in the wild in North Carolina. 15 That's
- 12:10terrifyingly low. It is, and the official red wolf
- 12:13recovery program has faced huge challenges, including
- 12:16hybridization with coyotes, which dilutes the red wolf
- 12:19genes. But didn't they find another
- 12:20population somewhere else? Sort of.
- 12:23They found wolves in Louisiana that actually had more red wolf
- 12:27DNA in them than the recognized population in North Carolina.
- 12:30It complicates the picture. So what did Colossal do?
- 12:33They successfully cloned 4 red wolves and importantly, they
- 12:37were from three different genetic lines.
- 12:40So increasing the genetic diversity which is crucial for
- 12:43such a small population. Precisely.
- 12:45And they developed a new cloning technique using blood samples
- 12:48endothelial progenitor cells. Non invasive.
- 12:51That sounds like a big deal for conservation.
- 12:53Huge makes biobanking genetic material much easier and less
- 12:57stressful for the animals. A potential game changer, as
- 13:00Lamb put it. But they hit roadblocks with
- 13:02getting more wolves cloned. Yeah, initially they offered to
- 13:05clone more for free, but apparently the US Fish and
- 13:07Wildlife Service was slow to respond or engage.
- 13:11Some frustration there. But things are looking up now.
- 13:14Seems so. He mentioned a recent positive
- 13:16meeting with the Department of the Interior, who seems
- 13:18supportive of their conservation work, so hopefully progress can
- 13:22be made. OK, let's get back to the
- 13:24direwolves, because they actually did.
- 13:26It they did, 3 direwolf pups have been born, named Romulus,
- 13:32Remus and Kesey. Wow, actual direwolf pups,
- 13:36that's incredible. It really is, Lam described
- 13:39Peter Jackson's reaction of, you know, the director.
- 13:42Lord of the Rings, he's involved.
- 13:44An investor? Yeah, Apparently Jackson heard
- 13:46the pups howl, the first direwolf howls in maybe 10,000
- 13:51years and got quite emotional. I bet and there was some weird
- 13:53coincidence with the Game of Thrones throne.
- 13:56Yeah, apparently the pups had a vet checkup scheduled and it
- 13:59happened to be near this actual prop throne from the show that
- 14:02Jackson had just acquired. Cosmic timing.
- 14:04That's pretty funny. Are the pups OK?
- 14:06Healthy? Seems so.
- 14:07They had the American Humane Society there, vet teams.
- 14:11Animal welfare is clearly a priority.
- 14:13They're not being released or anything.
- 14:14Where are they living? On a secure 2000 acre ecological
- 14:18preserve 247 care, they're being closely monitored.
- 14:23And they're growing fast. Incredibly fast, over 45 lbs at
- 14:27three months, 80 lbs by 5 months, and already 5.5 feet
- 14:32long. Oh, so bigger than Gray wolves.
- 14:34Definitely tracking that way. Fossil records show dire wolves
- 14:38were about 2025% larger and stockier than Gray wolves.
- 14:42These pups seem to be confirming that and.
- 14:44What about their appearance? Did the genetics reveal anything
- 14:47surprising? Yes, the genome apparently
- 14:50contains genes for white fur white.
- 14:52Not the reddish brown often depicted.
- 14:54Nope. So maybe some looked more like
- 14:55dosed from Game of Thrones. It challenges the old
- 14:58assumptions based on fossils alone.
- 15:00That's cool. Are they showing any direwolf
- 15:01behaviors yet? Early signs of hunting behavior.
- 15:04Apparently, even in captivity, those ancient instincts might be
- 15:07surfacing. So those 10,000 year old hunting
- 15:09genes, including the ones for size, are actually being
- 15:12reactivated. It seems like it.
- 15:14By comparing the DNA from that 13,000 year old tooth and the
- 15:1873,000 year old skull, they could pinpoint genes that were
- 15:22really fixed and conserved in the species.
- 15:25It's fascinating to think that these complex behaviors are just
- 15:29baked into the genes waiting to re emerge.
- 15:32Deeply fascinating, the idea of dormant characteristics coming
- 15:35back online after millennia. Are there other extinct animals
- 15:39Colossal is looking? At Definitely, the American
- 15:41Cheetah was mentioned. There was an American cheetah.
- 15:43Yep, now extinct but we have it's full genome.
- 15:46It's thought to be why pronghorns are so incredibly
- 15:49fast. They evolved to escape it.
- 15:50That makes sense. What else?
- 15:52The Stellar's sea cow, a huge, gentle marine mammal related to
- 15:56manatees. Oh, I've heard of that.
- 15:57Haunted to extinction really quickly.
- 15:59Right. Tragically fast.
- 16:00Within about 30 years of Europeans discovering it in the
- 16:03Bering Sea, we have its full genome too.
- 16:05And bringing it back could help ecosystems.
- 16:08Potentially, yeah. They were massive grazers of
- 16:11kelp. Restoring them could have big
- 16:13benefits for kelp forests, which are crucial marine habitats.
- 16:16Looking further ahead, what about technologies like
- 16:19artificial wombs? Is that part of the plan?
- 16:21It's on the horizon. Lamb sees it as a potential
- 16:24future tool for, as he put it, productionizing endangered
- 16:29species recovery. Meaning.
- 16:31Scaling up breeding efforts, increasing genetic diversity

- 19:23Wild dogs with a unique vocalization. 15 dogs, OK.
- 19:26And the elephant cancer resistance thing.
- 19:28Pedo's paradox. Elephants have way more cells
- 19:31than us and live a long time, so statistically they should get
- 19:34cancer much more often, but they don't much lower rates.
- 19:37They have extra copies of a tumor suppressor gene P53, which
- 19:41is cool but also makes gene editing and elephants tricky
- 19:44because the cells are so good at repairing DNA changes or self
- 19:48destructing if damaged. Anything else striking the?
- 19:52Cane toad resistance Some mammals in South America, where
- 19:55cane toads are native, have evolved resistance to their
- 19:58toxins. And the idea is to give that
- 20:00resistance to Australian animals.
- 20:02Potentially, yeah. Engineer that resistance into
- 20:05endangered Australian marsupials like quolls, which are
- 20:08devastated by invasive cane toads.
- 20:10Wow and giant Beavers pronghorn speed.
- 20:13Yeah, giant Beavers in place to see North America and the
- 20:16pronghorns incredible speed being an evolutionary echo of
- 20:19the extinct American cheetah. And that bird with claws?
- 20:23The Hodson, a South American bird whose chicks have claws on
- 20:26their wings, letting them climb trees before they can fly
- 20:29properly, looks very dinosaur like.
